LifeEZe SAM-e contains S-adenosylmethionine (SAM-e) 200mg in the form of sulfate/tosylate. Each tablet is enteric coated - Enteric coated tablets are less likely to breakdown in the stomach before reaching the intestine where it is primarily absorbed.

S-adenosylmethionine (SAM-e) is a physiological molecule occurring naturally in the tissues of the body, predominately in the liver, adrenal and pineal glands. An important metabolic intermediate, SAM-e is synthesised in vivo from ATP (adenosine triphosphate) and L-methionine by SAM-e synthetase to form a molecule composed of a purine (adenine: 6-aminopurine), a sugar (deoxyribose) and an amino acid (L-methionine).

SAM-e was first brought to attention by an Italian, Cantoni in 1951. It was an unstable molecule and could not be studied in clinical trials until it became commercially available in 1974. An Italian group devised a way of stabilizing S-adenosylmethionine by producing a derivative stabilized with p-toluene sulfate. Since then numerous studies have been carried out showing that SAMe participates in a variety of biological reactions.

SAM-e has been used clinically in Europe since the 1970's and is today the most prescribed natural supplement for stress, nervous tension, anxiety and low mood in Europe. "In the USA SAM-e became available in 1999 as a dietary supplement and is now the most purchased over-the-counter supplement in the United States!" Clinical studies using exogenous (oral or parenteral) SAM-e have found beneficial effects for those suffering from stress, nervous tension, anxiety, low mood, osteoarthritis, poor liver function as well as a number of other health conditions.

SAM-e has experimentally shown to be effective with those suffering from low mood disorders. SAM-e stimulates the activity of dopaminergic and serotonergic pathways in the brain through its involvement in the transmethylation processes. Levels of SAM-e synthetase and phosphatidylcholine have been shown to be reduced in the erythrocytes of patients with low mood suggesting a disruption in SAM-e pathways.

This has formed the basis for the use of SAM-e for the relief of stress and associated conditions, particular those suffering with a low mood. Clinical trials conducted have shown SAM-e to be as effective, in the low to moderate dosages, as drugs.

The beneficial effects of SAM-e began more quickly and produced fewer side effects. The effect of SAM-e in patients suffering from anxiety and associated health problems is seen by a consistent improvement in mood as determined by the Hamilton rating scale. Patients were given daily dosages of up to 1600mg daily with an onset of action within the first two weeks of treatment and no adverse effects noted.

SAM-e plays an important role in cartilage formation and repair. To date, a total of 22,261 patients have participated in clinical trials in the treatment for pain of osteoarthritis. 21,524 were given SAM-e, 458 were given different NSAIDs and 279 given placebo. The trials were conducted over a period of three weeks to two years and dosages ranged from 400 - 1200mg daily. Data from the controlled trials have shown SAM-e's superiority to placebo and a similar degree of efficacy as NSAIDs without side effects when compared long term.

Unlike NSAIDs' the current choice of treatment for the relief of pain for osteoarthritis, SAM-e does not inhibit prostaglandin synthesis, does not cause gastrointestinal damage and does not inhibit platelet aggregation. Instead, SAM-e promotes in vitro synthesis of proteoglycans by human chondrocytes, is free from systemic side effects and exhibits clinical effectiveness.

SAM-e has shown hepatoprotective effects in clinical trials. In patients presenting with liver problems such as metabolism of methionine, transmethylation and transsulphuration pathways are impaired reducing the activity of SAM-e synthetase, the enzyme required for synthesizing SAM-e from ATP and L-methionine.

S-adenosylmethionine is able to increase the intracellular glutathionine (the main cellular anti-oxidant) levels in the liver by activating the trans-sulphuration pathway, which is known to be low in those who may have sluggish liver rendering it effective in this area.
